WUSM, the Department of Neurology, and its associated Alzheimer's Disease Research Center has wellestablished research programs and a renowned group of faculty committed to research, education, and training. We hypothesize that at least some of these risk factors are genetic and that they may also modify amyloid deposition. The aim of this proposal is to use a genetic linkage strategy to identify new genetic risk factors for late onset AD. As our test sample we will use three hundred caucasian sib pairs with an age of onset of AD over the age of sixty five years to perform a 20cM genome-wide screen using microsatellite markers. Genomic regions showing evidence of linkage will be followed up with flanking markers in the same sample and in a second sample of equivalent size also selected on the basis of racial origin and age of onset.
